Compared with the
traditional low-pressure gas discharge plasma source, atmospheric pressure room
temperature plasma (ARTP) has the characteristics of low plasma jet temperature, uniform discharge and high concentration of chemically active
particles. Based on ARTP technology, Our company, together with the Department
of chemical engineering and the Department of Engineering Physics of Tsinghua
University, has jointly developed the first special instrument for mutation and
breeding of microorganisms by means of plasma — ARTP Mutagenesis Breeding Machine. The instrument has high mutation
rate, compact structure, simple operation, high safety and fast mutation speed.
A large mutation library can be obtained by one mutation operation (within a
few minutes), which greatly improves the intensity of strain mutation and the
capacity of mutation library; ARTP technology combined with high-throughput
screening technology can realize rapid and efficient evolutionary breeding of
organisms.

Case 1：Screening of High Salt Tolerant Mutant Strains by ARTP
"After plasma treatment, when Enterobacter cloacae was cultured in the medium containing 7.5% NaCl, a salt tolerant mutant appeared, and the degradation percentage of TPH was 2.5 times higher than that of the original strain. Moreover, when the mutant was cultured under the condition of LB + 9.0% NaCl, it could quickly accumulate k+ in the cell and EPS outside the cell."

Case 2： High Throughput Screening of Mutant Strains of Oil Producing Yeast by ARTP Combined with 96 Well Plate
ARTP mutation breeding method can quickly and effectively mutate the oil producing yeast rhodosporidium toruloides with good effect. Through the screening of 44 mutagenesis strains, the mutation rate was 47.2%, and the positive mutation rate was 27.2%. A high-throughput screening method based on Nile red staining was established to screen a mutant strain(C4) of high oil and fat, which was cultured in YEPD culture medium for 78h, and its oil production was 2.2 times that of the original strain, and its biomass was increased to 1.5 times that of the original strain.
